Subject: adjusting the shifter, newbie question

Greetings all
I've removed the battery tray on my '72 ambo to get to the upper split
bolt wich was worn and needed replacement.
While at there, I thought I could adjust the shifter mechanism by
loosening the locknut and rotate the adjusting screw according to
guzziology by 1/4 turn at the time.
Another book (chilton's tune up guide) says to adjust until the gears
shifts smoothly throughout the range.
My newbie question is: Is that, adjusting a little bit and then
staticaly go trough the gears, until they are smooth. Or do I reassamble
the lot, and take the bike for a ride to test this??
I kind'a hope it's a static test, otherwise it would be tedious!
